The CHARIOT PRO Substudy (CPSS) was initiated in 2015 as an industry sponsored (Johnson & Johnson) prospective longitudinal study, at two Sites in the United Kingdom. Imperial College London (ICL n=409) and the University of Edinburgh (UEI, n=112), led by Lefkos Middleton MD, of Imperial College London. The study’s primary goal was to evaluate factors of risk and/or protection for cognitive decline and dementia due to Alzheimer’s disease (AD) and related neurodegenerative diseases. Towards this goal, the study employed magnetic resonance imaging (MRI), positron emission tomography (PET) and fluid-based biomarkers (cerebrospinal fluid and blood-based) as well as repetitive clinical and multiple neuropsychological assessments to track cognitive trajectories longitudinally. The UEI site was terminated in 2000; hence, the complete cognitive trajectories to study end and all fluid biomarker measurements were derived and conducted at ICL only. Additional funding from Merck, Takeda and Gates Ventures allowed, in 2022, a two- year extension of CPSS, enriched with one structural and functional MRI, followed by up to three Tau PET scans, at ICL.
