Plasma biomarkers and cognitive measures in Chinese Americans with Alzheimer’s disease and mild cognitive impairment

Description:
Thirty-four Chinese American participants were included in this retrospective study, recruited through the Alzheimer’s Disease Research Center (ADRC) at the University of California, San Francisco (UCSF) Memory and Aging Center (MAC). The cohort comprised n = 19 clinically normal, n = 3 with mild cognitive impairment, and n = 12 Alzheimer’s disease participants. Upon enrollment, all participants underwent a clinical examination including a battery for neuropsychological evaluation, health history, examination by a behavioral neurologist, and participated in biospecimen collection for laboratory testing. Plasma biomarker data. Whole blood was collected in two 10 ml LLT Plasma tubes and centrifuged within 1 hour of collection at 4°C for 10 minutes at 2000xg. Samples were immediately frozen at −80°C as 0.5ml aliquots in cryopreservation tubes and stored until use. Plasma biomarker data was obtained on a MESO QuickPlex SQ 120 Imager using Discovery Workbench v4.0 to analyze the results. Vascular and immune-related plasma biomarkers were run in either a single, 2-plex, or 3-plex assay format, utilizing individual kit guidelines per manufacturer protocol. All samples and standards were run in duplicate. Cognitive testing data. English or Chinese language versions of the Cognitive Abilities Screening Instrument and the UCSF MAC Bedside Neuropsychological Screen were used to assess composite memory, executive function, visual construction, and language performance for each participant. Cognitive domain aggregates used in analysis were (i) aggregated by summing individual test scores comprising respective domains with agreeable scale distributions prior to (ii) Z-scoring relative to the cognitively normal group. Tests with unique score scale distributions were first Z-scored before being averaged in to respective domain aggregates. Data processing and analysis. Pearson correlation analysis was first conducted including all biomarkers and cognitive testing measures for identification of broader relationships between peripheral and cognitive measures. An angiogenesis composite score based on initial tests was created by averaging individual Z-scores from MMP-1, bFGF, VEGF, and VEGF-C to further explore relationships after correcting for covariates (diagnosis, age, sex, APOE status, education) in a multiple linear regression analysis. All statistical analyses were performed in R.
