Neuropathological quantifications, transcriptome, and in situ proteomics profile datasets for neuronal basis of circadian rhythm dysfunction in tauopa

Description:
We conducted comprehensive analyses on patients with Alzheimer"s (AD) and Progressive Supranuclear Palsy (PSP), as well as healthy controls, focusing on specific brain nuclei related to sleep and wake regulation and control regions. The selected areas include the locus coeruleus (LC), lateral hypothalamic area (LHA), tuberomammillary nucleus (TMN), suprachiasmatic nucleus (SCN), supraoptic nucleus (SON), and paraventricular nucleus of the hypothalamus (PVN). Participants were categorized into Health Controls (with no clinical diagnosis of tauopathy and Braak Stage <= 2), AD patients, and PSP patients. For RNA-level expression analysis, we employed NanoString nCounter technology to measure RNA levels in the LC, LHA, and TMN. Our Neuropathology gene panel, tailored to encompass circadian rhythm genes from the transcription-translated feedback loop (TTFL) machinery, was used (details on the panel composition and gene list are available in RFL files). In terms of in-situ proteomic profiling, we utilized NanoString Digital Spatial Profile technology to assess protein expression within Areas of Interest (AoI) located within the SCN, SON, and PVN nuclei. These AoIs were identified using AVP, VIP, and GFAP protein markers, and we measured 53 proteins of interest (the protein list can be found in XLSX files together with normalized protein level data).
